dc.description.abstract | The objective of this research was to test the sensitivity of in vitro matured bovine oocytes to the addition of the cryoprotectant, DMSO, at room temperature and at O C. In the first experiment, oocytes were randomly assigned to either a 5, 10, 15, 20, or 30 min exposure to 1.5M DMSO in Dulbecco's phosphate-buffered saline at room temperature (27 C). Normal fertilization rates were found to be significantly different from controls in the 5, 20 and 3 0 min exposure times (P < 0. 05). Normal fertilization rates in the IO and 15 min exposure times were not significantly different from the controls or each other. In the second experiment, oocytes were exposed to 1.5M DMSO at 0 C for either 15, 20, or 30 min exposure times. Normal fertilization rate was found to be significantly higher in the 15 min exposure to DMSO when compared to the 20 or 30 min times and to the 20 min control. No results were found to support the theory that exposing oocytes to cryoprotectant at O'C rather than 27'C lessens the toxicity of the cryoprotectant. | en |
