dc.description.abstractThis study examined the response of bovine CD4⁺, CD8⁺, and TCR γ[] T lymphocytes to activation with the mitogens concanavalin A (Con A), phytohemagglutinin (PHA), and pokeweed mitogen (PWM), and the superantigens Staphylococcus enterotoxin A (SEA) and SEB. Several molecules (CD25, CD71, ACT1, PCNA, cyclin D, cdk4, and cdk6) were tested for suitability as activation markers. The marker of activation chosen was CD25, the interleukin-2 receptor α chain. Cells were analyzed by flow cytometry, and the mean fluorescence intensity ratio was determined. A dose response was found to each activating agent in all 3 T cell subsets. A relationship between time in culture with the activating agent and CD25 expression was found to exist in CD4⁺ and TCR []γ cells, but not CD8⁺ cells. This study documents, for the first time, mitogen- and superantigen-induced activation of bovine T cell subsets at early time points.en
