Abstract

Horizontal starch gel electrophoresis was used to examine protein variation in five Macrobrachium species. The hemolymph proteins hemocyanin, general protein, and esterase were compared in four North American species (M. acanthurus, M. carcinus, M. ohione, and M. olfersii). Eight proteins from homogenates of tail muscle and cephalothorax tissue also were examined in the North American species and in M. rosenbergii. The three hemolymph proteins varied interspecifically but no intraspecific variation was identified for M. acanthurus, M. carcinus, and M. ohione. The hemolymph proteins investigated in this study were not promising as genetic markers. Some nongenetic protein variation was noted, but it could not be determined if this was related to reproductive conditions, moulting stages, or possibly from the electrophoretic conditions. Hemolymph esterase could not be resolved clearly. Several esterase substrates and dye couples were tested. Six M. ohione geographic populations were examined for 9 proteins and 14 loci. Estimates of proportion of polymorphic loci, mean heterozygosity per locus, and mean heterozygosity per individual ranged from 21.4 to 42.8%, 12.9 to 24.1%, and 12.9 to 24.1% respectively. Estimates of genetic similarity and genetic distance also were calculated and all the populations were essentially genetically similar. Significant differences were found between allele frequencies for some of the pair combinations between M. ohione geographic populations. No significant differences were detected between allele frequencies when the M. ohione allele frequency data were pooled and recalculated according to sex.
