dc.description.abstractLeaf tissue derived from a systemic host N. tabacum L. var. Samsun and a local lesion host N. tabacum I. var. Samsun NN of tobacco mosaic virus (TMV) was cultured in a modified Murashige and Skoog's liquid medium (M&S). Cell chains were isolated eight to fifteen days after culturing and subsequently sub-cultured in Leitz micro-culture dishes containing 25 μl of conditioned M&S medium surrounded by mineral oil. The cell chains had four to fourteen cells per chain. Two cell chains were placed in each micro-culture dish and one cell of one chain was micro-injected with either infectious or heat-treated TMV, and the other served as an uninjected control. Cells were injected with the aid of heat drawn glass needles, mounted in a micro-manipulator. Forty eight Samsun NN cell chains were injected. Twenty four injected cell chains were lost to injection damage and culture problems, nine chains became infected, two chains showed no symptoms and/or sign of infection, three cell chains were injected with heat-treated virus, and ten were used for other purposes. Twenty two cell chains of the Samsun variety were injected. All ten cell chains injected with infectious TMV became infected, five were injected with heat-treated TMV, and seven were lost or used for other purposes..en
