Scoring and mapping bovine anchor loci and screening polymorphic markers for horns and coat color in a Bos indicus X Bos taurus cross
Abstract
Backcross and F2 families from a cross between Bos taurus (Angus) and Bos indicus (Brahman) have been scored for DNA markers in the Mechanisms of Genetic Control of Beef Carcass Merit Traits project (the "Angleton project") in an effort to locate genes influencing 56 qualitative and quantitative traits. Fifty five microsatellite markers were tested for use in the quantitative trait locus (QTL) search. The microsatellite markers are distributed as follows: 24 markers are assigned to bovine chromosomes or syntenic groups, 12 markers are assigned to linkage groups, and 19 are unassigned and unlinked markers. In order to more efficiently assign the markers under development to chromosomes, additional anchor loci were scored and tested for association with these markers. Once a genetic map is available, associations with phenotypic traits can be analyzed and genes involved in the expression of these traits can be localized to a chromosomal region flanked by adjacent markers. Several protein polymorphisms have been mapped to bovine chromosomes and were selected for scoring in the Angleton families. Ten blood group systems, the blood proteins hemoglobin, albumin, transferrin and the liver enzymes: alcohol dehydrogenase, glucose-6-phosphate isomerase, glutamate oxaloacetate transaminase, lactic acid dehydrogenase, malate dehydrogenase, malic enzyme, mannose-6-phosphate isomerase, nucleoside phosphorylase, 6-phosphogluconate dehydrogenase, and phosphoglucomutase 2 were assayed in the Angleton families with the goal of mapping these markers to eleven chromosomes and 19 syntenic groups. The protein markers were tested for association with three phenotypes and the microsatellite markers scored in the Angleton families. Eleven of the protein systems-were informative as anchor loci and were found to be located on eight different chromosomes and one syntenic group. The hom-polled and the black-nonblack traits were found to be associated with genetic markers. The speckled-nonspeckled trait was not found to be associated with any of the markers screened in the Angleton families. These 3 traits, which have been reported to be simple qualitative traits, proved to be "pressed as qualitative traits influenced by a major gene and with varying degrees of penetrance.
